Pinealon (EDR peptide, Glu-Asp-Arg) is a synthetic tripeptide from the Khavinson bioregulator family, studied in neuroprotection, brain-aging, and circadian-biology research. Published work in neuronal cell models reports reduced reactive-oxygen-species accumulation and improved cell viability under oxidative stress.

Research Overview

The Khavinson tripeptide suddenly at the center of the sleep-and-longevity conversation

Pinealon is a synthetic three-amino-acid peptide (Glu-Asp-Arg, 'EDR') from the family of short 'peptide bioregulators' developed by Vladimir Khavinson's group at the St. Petersburg Institute of Bioregulation and Gerontology. Designed with the pineal gland and brain aging in mind, it spent decades in relative obscurity — until it abruptly became one of the most-searched peptides of 2026.

Neuroprotection under oxidative stress

Preclinical (cell & animal)

The best-documented Pinealon finding: in neuronal cell models, Pinealon dose-dependently limited reactive-oxygen-species accumulation and reduced cell death under oxidative stress, while supporting proliferative activity.

Brain-aging & Alzheimer's models

Preclinical (cell & animal)

In an in vitro Alzheimer's-model preparation, EDR and related short peptides restored dendritic spine density; follow-on work reported protective effects in aged, fibroblast-derived induced neurons.

Proposed epigenetic mechanism

Preclinical (cell & animal)

Khavinson's group hypothesizes that short peptides like EDR penetrate cells and interact directly with DNA to modulate gene expression and protein synthesis — a provocative mechanism explored in the context of Alzheimer's pathogenesis, but not yet independently confirmed.

Sleep & circadian biology

Preclinical (cell & animal)

Pinealon is named for the pineal gland, the brain's melatonin factory, and sits within a research program on pineal aging. Direct evidence that Pinealon improves human sleep, however, is currently anecdote — including the high-profile anecdote that made it famous. It is the single biggest open question about this compound.

Nearly the entire Pinealon literature comes from one research group (Khavinson and collaborators), largely in Russian journals. Independent replication is scarce.
